In an article published as the cover story of the March 2013 issue of Nature Chemical Biology, Lindsey James, PhD, research assistant professor in the lab of Stephen Frye, Fred Eshelman Distinguished Professor in the UNC School of Pharmacy and member of the UNC Lineberger Comprehensive Cancer Center, announced the discovery of a chemical probe that can be used to investigate the L3MBTL3 methyl-lysine reader domain. The probe, named UNC1215, will provide researchers with a powerful tool to investigate the function of malignant brain tumor (MBT) domain proteins in biology and disease.
“Before this there were no known chemical probes for the more than 200 domains in the human genome that recognize methyl lysine. In that regard, it is a first in class compound. The goal is to use the chemical probe to understand the biology of the proteins that it targets,” said Dr. James.
Chromatin regulatory pathways play a fundamental role in gene expression and disease development, especially in the case of cancer. While many chemical probes work through the inhibition of enzyme activity, L3MBTL3 functions as a mediator of protein-to-protein interactions, which have been historically difficult to target with small, drug-like molecules.The researchers found three to four further disease subtypes within TN tumors, with more than 75 percent of the tumors falling into the basal-like subtype. Further research is needed to identify the distinct biomarkers shared by the expanded subtypes of TN cancers. The ultimate goal will be to target the individual biomarkers of these subtypes and create therapies that target their individual biology, according to Dr. Perou.
“Many people believe that protein-protein interactions are difficult to target. Often they have a large surface area, so it is hard for small molecules to go in and intervene,” said Dr. James.
Almost 40 percent of the genes that drive cancer can be mapped to dysfunction within signaling pathways. In the last five years, chemical probe development has allowed researchers to make fundamental observations of the role of these pathways in cancer development, as well as pointing to potential targets for new therapies. Each of the complex interactions within the signaling pathways represents a potential point where a therapy can be applied, and the probes allow researchers to interact with these processes at the molecular level and observe the overall effect of their perturbation on the disease state.
In a 2008 Nature Chemical Biology commentary, Dr. Frye outlined the qualities that make a good chemical probe. To Frye, a good chemical probe must be highly selective to enable specific questions to be asked and it must function as well in a cell as in the test tube, providing clear quantitative data with a well understood mechanism of action in either situation. It also must be available to all academic researchers without restrictions on its use, a criteria that the L3MBTL3 probe fulfills through the Frye lab’s commitment to provide researchers with the probe free of charge on request and UNC1215 is already available through commercial vendors as well.

though called “turkish” paper marbling by europeans, this design technique was developed in east asia, central asia, and the islamic world. it is an important part of turkic, tajik, indian, and other asian and middle eastern cultures.

Tolkien was able to draw.

When J.R.R. Tolkien wrote The Hobbit, he was already an accomplished amateur artist, and drew illustrations for his book while it was still in manuscript. The Hobbit as first printed had ten black-and-white pictures, two maps, and binding and dust jacket designs by its author. Later, Tolkien also painted five scenes for color plates, which comprise some of his best work. His illustrations for The Hobbit add an extra dimension to that remarkable book, and have long influenced how readers imagine Bilbo Baggins and his world.

I have found The Art of The Hobbit book here in Amazon. Some of these images are published here for the first time, others for the first time in color, allowing Tolkien’s Hobbit pictures to be seen completely and more vividly than ever before.

Hecate- Goddess of the Night and Magic

Her name roughly translates as “Worker from afar”, she is a particularly mysterious other world divine being in the Greek pantheon. Hecate was seen as the goddess of magic, witchcraft, night, moon, ghosts and necromancy.

She was the only child of the titans Perses and Asteria from whom she received her powers over the heavens, earth and seas. She could bestow mortals with wealth, victory, wisdom, good luck to sailors and hunters, and prosperity to youth and to herds of cattle. She could also withhold these gifts. After the defeat of the Titans, Zeus honoured Hecate and allowed her to maintain her dominion.

She is usually depicted as a woman holding twin torches on Greek pottery. She sometimes wears a knee length maiden skirt and hunting boots, much like the goddess Artemis. In statues Hekate is often depicted in a curious triple form as goddess of crossroads.

Her familiars were a black she-dog and a polecat. The black dog was said to be the Trojan queen Hekabe who leapt intot he sea after the fall of Troy and was transformed into a dog by the goddess. The polecat was said to have originally been a witch called Gale who was transformed into a polecat as punishment for her incontinence. Others told that the polecat was Galinthias, nurse of Alkmene, transformed by the angry Eileithyia, but received kindly by Hecate as her animal.

She aided Demeter in her search for Persephone using her torches to light the goddess’ way throughout the darkness of night. After Persephone’s return Hecate became her companion and minister in Hades.

She was identified with a number of other goddesses in the ancient world which lead to some confusion which lasts to this day. These include Artemis the Virgin Huntress, Selene the Moon goddess, the sea goddess Krataeis, the Thracian goddess Bendis, Maira the goddess of the Dog Star, and the Eleusinian Daeira.

Her titles include: the destroyer (Perseis), angry/terrible one (Brimo), lady of the underworld (Aidonaia), three formed/three bodied (Trimorphis), of the crossroads (Trioditis), night wandering (Nyktipolos), tender/delicate (Atalos), nurse of the young (Kourotrophos), leader of dogs (Skylakagetis), and queen of those below (Anassa Eneroi).

 She had very few public temples in the ancient Greek world however. Instead she was honoured at small household shrines. These were probably made to help ward off evil and malevolent powers of witchcraft. Her most important cultic sites were at Eleusis, and Samothrace. The mysteries of Samothrace were celebrated in Hecate’s honour.

This is extremely true from what I’ve heard. MY school does some life drawing with clothing, usually a student, and I plan on taking a few classes with nude models in a local gallery.
